Eteplirsen je prvi lek u terapiji DMD iz grupe novih terapija takozvanog preskakanja egzona (eng. exon skipping) koji je registrovala američka Uprava za hranu i lekove (Food and Drug Administration – FDA). Ovaj vid terapije nije pogodan za sve pacijente obolele od DMD, već je rezervisan za one koji imaju specifičan tip mutacije koja remeti okvir čitanja, u konkretnom slučaju pogodne za preskakanje egzona 51 u DMD genu, što čini oko 14% svih pacijenata. [15] Efekat preskakanja egzona se postiže primenom sintetičkih analoga nukleinske kiseline zvanih „antisens” – besmislenih oligonukleotida (antisense oligonucleotide – AON). Uloga AON jeste vezivanje za komplementarnu sekvencu pre-iRNK, u ovom slučaju egzon 51, vršeći uticaj na iskrajanje egzona na način da se određeni, u ovom slučaju egzon 51 ne nađe u finalnom transkriptu, što koriguje translacioni okvir čitanja i omogućava sintezu skraćenog, ali funkcionalnog proteina distrofina (Slika 3).“

"1-methyl-2-pyrrolidinone (NMP; 20 mL/g resin) is added to aminomethylpolystyrene resin (100-200 mesh; ˜1.0 mmol/g load based on nitrogen substitution; 75 g, 1 eq, Polymer Labs, UK, part #1464-X799) in a silanized, jacketed peptide vessel and the resin is allowed to swell with mixing for 1-2 hr. Following evacuation of the swell solvent, the resin is washed with dichloromethane (2×1-2 min), 5% diisopropylethylamine in 25% isopropanol/dichloromethane (2×3-4 min) and dichloromethane (2×1-2 min). After evacuation of the final wash, the resin is treated with a solution of disulfide anchor 34 in 1-methyl-2-pyrrolidinone (0.17 M; 15 mL/g resin, ˜2.5 eq) and the resin/reagent mixture is heated at 45° C. for 60 hr."
