Lilly signaled a ***slight imbalance of thromboembolic events*** in baricitinib studies, causing the FDA to request additional data ...
The thromboembolic events for baricitinib were 0.46 per 100 years, which appear reasonable as the typical range for the patient group is 0.3-0.8. We are surprised by the FDA request given the drug’s safe profile and prior European and Japanese approvals. We expect additional clinical data and real-world use outside the U.S. will lead to eventual U.S. approval by 2019.

"The FDA has indicated that a new clinical study is necessary ... in light of the observed imbalance in thromboembolic events that occurred during the placebo-controlled period of the RA clinical program.
... Thromboembolic events - diagnosed as deep venous thrombosis (DVT) and pulmonary embolism (PE) - were reported in five patients receiving baricitinib during the controlled period of two of seven completed Phase 2 or Phase 3 trials in RA. Although an imbalance was observed during the placebo controlled period of the RA clinical trials, the rate of these events in the overall baricitinib clinical program was consistent with that seen among the general population of treated RA patients."
