Friedreich’s ataxia (FA) is a disease caused by a reduction in the levels of a protein called frataxin. What is known about the cause of FA is that the low levels of frataxin are due to a mutation in the gene coding for frataxin (FRDA), in particular a 3-base pair sequence (GAA) repeated up to 1700 times in the mutated FRDA gene. The way this mutation affects frataxin levels is not completely clear although several hypotheses have been described. In spite of several therapeutic strategies being under development, currently there is no treatment for Friedreich’s ataxia.

The human cell model developed in our laboratory represents a novel tool which will help us understand more about the mechanism through which the GAA mutation affects the levels of the protein frataxin, causing FA. Our aim is to use these cells as a platform for the screening and identification of novel therapies for Friedreich’s ataxia, namely molecules able to increase frataxin cellular levels.

L’atassia di Friedreich (AF) è una malattia causata dalla riduzione dei livelli di una proteina denominata fratassina. Ciò che si conosce sulla causa dell’AF è che questi livelli bassi di fratassina sono dovuti a una mutazione nel gene che codifica per la fratassina (FRDA), in particolare una sequenza di 3 basi (GAA) che può ripetersi fino a 1700 volte nel gene mutato. Il modo in cui questa mutazione influenza i livelli di fratassina non è ancora del tutto chiaro, nonostante vi siano varie ipotesi a riguardo. Il modello cellulare umano sviluppato nel nostro laboratorio rappresenta uno strumento innovativo che ci permetterà di comprendere meglio il meccanismo attraverso il quale la mutazione GAA influenza i livelli di fratassina, causando l’AF.
Il nostro obiettivo è utilizzare queste cellule come piattaforma per lo screening e l’identificazione di nuove terapie per l’atassia di Friedreich, nella fattispecie molecole capaci di aumentare i livelli di fratassina nelle cellule.
