Translation - Chinese Phosphatase and tensin homolog (PTEN, once named phosphatase and tensin homolog deleted on chromosome ten) is a newly identified tumor suppressor gene which has been shown to play important roles not only in cell proliferation, differentiation and apoptosis, but also potentially in the regulation of cell growth, tumor infiltration, metastasis and angiogenesis [4,5].
Recent studies have revealed several putative mechanisms relating to tumor suppression. First, PTEN inhibits the phosphatidylinositol-3-phosphate (PIP3) pathway by dephosphorylating PIP3, ultimately preventing cell growth and promoting cell apoptosis [4,5]. Correspondingly, inactivation of the PTEN gene can result in over-proliferation of cells and decreased apoptosis, facilitating tumor growth and transformation.
Second, PTEN can also significantly inhibit adhesions, facilitate migration and spread of a variety of tumor cells, thus closely linking PTEN loss to tumor invasion and metastasis [7]. Studies have shown that PTEN inhibits cell infiltration and metastasis by dephosphorylating focal adhesion kinase (FAK) which plays critical roles in tumor invasion and metastasis, thus inhibiting cell proliferation, cell adhesion, migration, and local spread [8]. Both in vitro and in vivo studies have shown FAK dephosphorylation by PTEN with PTEN overexpression consequentially inhibiting integrin-mediated cell expansion, focal adhesion formation, and cell migration. The findings are supported by antisense study in which knockdown of PTEN enhanced cell migration. Additionally, FAK overexpression antagonized the effects of inhibition of PTEN on tumor cell infiltration and metastasis. Several groups [4,5,10] observed that, in liver cancer cells SMMC-7721 treated with TGF-β1 (transforming growth factor beta 1), integrin α5β1 expression was significantly elevated while PTEN protein expression was significantly decreased, which led to dramatically enhanced phosphorylation of FAK, and enhanced adhesion and migration to fibronectin/laminin of the extracellular matrix.
A study led by Jinyi Li et al. suggests that PTEN gene inactivation correlates with the occurrence of many human malignancies [6]. In non-cancerous mucosa, early gastric cancer, and advanced gastric cancer, PTEN were detected as 76.3% , 45.5% and 34.2% at mRNA level and 100%, 63.6% and 47.4% at protein level, respectively, suggesting abnormal expression of the PTEN gene was involved in the malignant transformation of gastric epithelial cells.
